RESUMO

To identify Janus kinase (JAK) inhibitors that selectively target gastrointestinal tissues with limited systemic exposures, a class of imidazopyrrolopyridines with a range of physical properties was prepared and evaluated. We identified compounds with low intrinsic permeability and determined a correlation between permeability and physicochemical properties, clogP and tPSA, for a subset of compounds. This low intrinsic permeability translated into compounds displaying high colonic exposure and low systemic exposure after oral dosing at 25 mg/kg in mouse. In a mouse PK/PD model, oral dosing of lead compound 2 demonstrated dose-dependent inhibition of pSTAT phosphorylation in colonic explants post-oral dose but low systemic exposure and no measurable systemic pharmacodynamic activity. We thus demonstrate the utility of JAK inhibitors with low intrinsic permeability as a feasible approach to develop gut-restricted, pharmacologically active molecules with a potential advantage over systemically available compounds that are limited by systemic on-target adverse events.

RESUMO

A prevalent observation in high-throughput screening and drug discovery programs is the inhibition of protein function by small-molecule compound aggregation. Here, we present the X-ray structural description of aggregation-based inhibition of a protein-protein interaction involving tumor necrosis factor α (TNFα). An ordered conglomerate of an aggregating small-molecule inhibitor (JNJ525) induces a quaternary structure switch of TNFα that inhibits the protein-protein interaction between TNFα and TNFα receptors. SPD-304 may employ a similar mechanism of inhibition.

Leukotrienes (LTs) and related species are proinflammatory lipid mediators derived from arachidonic acid (AA) that have pathological roles in autoimmune and inflammatory conditions, cardiovascular diseases, and cancer. 5-Lipoxygenase activating protein (FLAP) plays a critical accessory role in the conversion of AA to LTA4, and its subsequent conversion to LTC4 by LTC4 synthase. Pharmacological inhibition of FLAP results in a loss of LT production by preventing the biosynthesis of both LTB4 and LTC4, making it an attractive target for the treatment of inflammatory diseases in which LTs likely play a role. Small-molecule (SM) drugs often exhibit polypharmacology through various pathways, which may explain the differential therapeutic efficacies of compounds sharing structural similarity. We have profiled a series of SM FLAP modulators for their selectivity across enzymes of AA cascade in human whole blood (HWB), using a recently developed LC/MS (liquid chromatography-mass spectrometry)-based high-throughput lipidomics platform that monitors 122 eicosanoids in multiplex. Highly efficient data acquisition coupled with fast and accurate data analysis allowed facile compound profiling from ex vivo study samples. This platform allowed us to quantitatively map the effects of those SMs on the entire AA cascade, demonstrating its potential to discriminate structurally related compounds.

An enantioselective synthesis of the maoecrystal V (1) carbon skeleton is described. The key transformations include arylation of a 1,3-dicarbonyl compound with a triarylbismuth(V) dichloride species, oxidative dearomatization of a phenol, and a subsequent intramolecular Diels-Alder reaction.

The synthesis and electrochemical and photophysical studies of a series of alkyne-linked zinc-porphyrin-[60]fullerene dyads are described. These dyads represent a new class of fully conjugated donor-acceptor systems. An alkynyl-fullerene synthon was synthesized by a nucleophilic addition reaction, and was then oxidatively coupled with a series of alkynyl tetra-aryl zinc-porphyrins with 1-3 alkyne units. Cyclic and differential pulse voltammetry studies confirmed that the porphyrin and fullerene are electronically coupled and that the degree of electronic interaction decreases with increasing length of the alkyne bridge. In toluene, energy transfer from the excited zinc-porphyrin singlet to the fullerene moiety occurs, affording fullerene triplet quantum yields of greater than 90 %. These dyads exhibit very rapid photoinduced electron transfer in tetrahydrofuran (THF) and benzonitrile (PhCN), which is consistent with normal Marcus behavior. Slower rates for charge recombination in THF versus PhCN clearly indicate that charge-recombination events are occurring in the Marcus inverted region. Exceptionally small attenuation factors (beta) of 0.06+/-0.005 A(-1) demonstrate that the triple bond is an effective mediator of electronic interaction in zinc-porphyrin-alkyne-fullerene molecular wires.
